Oncostatin M (OSM) is a multifunctional, secreted cytokine that belongs to the IL-6 family. It signals through two main receptor complexes, Type I (gp130 + LIFR) and Type II (gp130 + OSMRβ), activating several intracellular signaling pathways including JAK/STAT, RAS/MAPK, JNK/p38 MAPK, and PI3K/AKT. OSM exhibits pleiotropic effects across multiple physiological systems, including regulating cell proliferation, differentiation, apoptosis, modulating immune responses, and influencing hematopoiesis. Elevated levels or dysregulation of OSM are implicated in several diseases, including rheumatoid arthritis, pulmonary inflammation/fibrosis, and cardiovascular diseases. Therapeutic strategies targeting the OSM signaling axis, such as monoclonal antibodies and small molecule inhibitors, are under investigation.
