Overview

Open reading frame 3a protein (ORF3a) is an accessory protein encoded by the genome of severe acute respiratory syndrome-associated coronaviruses, including SARS-CoV and SARS-CoV-2. It is the largest accessory protein in these viruses and plays significant roles in virus-host interactions, although it is not essential for viral replication in vitro or in vivo. ORF3a contributes significantly to pathogenesis by enhancing virus-induced cytopathic effects, altering intracellular trafficking and modulating immune responses indirectly through effects on cell death pathways. It is a homo-tetramer with each monomer containing three transmembrane domains. The tetrameric structure creates hydrophobic cavities that may be involved in ion channel activity ("viroporin" function) and interactions with host proteins.
