The opioid growth factor receptor (OGFr) is a unique receptor that specifically binds the endogenous opioid peptide [Met(5)]-enkephalin, also known as opioid growth factor (OGF). It is not structurally related to classical opioid receptors and mediates growth-inhibitory effects through direct modulation of gene expression after translocation into the nucleus. It plays critical roles in embryonic development, wound healing, cellular renewal, angiogenesis, cancer biology, and tissue repair processes. Modulating this axis holds promise for cancer management and autoimmune disorders.
Increases synthesis of cyclin-dependent kinase inhibitors p12 and p16, leading to inhibition of retinoblastoma protein phosphorylation, causing cell cycle arrest at G1 phase.
