Overview

Opioid growth factor receptor like 1 (OGFRL1) is a protein-coding gene structurally homologous to the opioid growth factor receptor (OGFR), thought to act as a potential orphan receptor with putative signaling activity and largely unknown biological roles[1][3][4]. OGFRL1 is highly expressed in myeloid lineage cells and tissues such as bone marrow, brain, and spleen[1]. Its function remains poorly elucidated, but pathogenic homozygous mutations in OGFRL1 have recently been linked to autosomal recessive cherubism, a severe craniofacial disorder presenting as jawbone expansion in children[1]. Unlike OGFR, which mediates effects of opioid peptide growth factor (Met5-enkephalin) and is involved in tissue homeostasis and immune modulation, the role of OGFRL1 in physiologic and pathologic signaling is still being determined. Some evidence indicates OGFRL1 may be involved in skeletal muscle differentiation and may interact with cell cycle regulators such as CDK4[4]. No evidence currently supports OGFRL1 as a drug target, nor are any drugs or clinical biomarkers associated with OGFRL1. Further investigation of OGFRL1’s function—especially in human craniofacial and immune processes—is warranted[1][3][4].
