The kappa opioid receptor (KOR) is a G protein-coupled receptor that binds to dynorphins and modulates several physiological processes including analgesia, stress response, mood regulation, reward processing, and inflammation control. Activation can induce diuresis and cause dysphoria and aversion. Unlike mu-opioid receptors, it does not cause respiratory depression, making it a potentially safer target for pain management. Widespread clinical use is limited by adverse effects such as dysphoria/sedation/aversion.
Agonist binding leads to Gi/G0 protein coupling, inhibition of adenylyl cyclase, and reduced neuronal excitability.
