Overview

ORC ubiquitin ligase 1 (OBI1) is an essential E3 ubiquitin ligase in humans, involved in selecting origins of DNA replication and activating them via multi-mono-ubiquitination of ORC3 and ORC5 subunits during S phase. It is a chromatin-associated factor whose activity ensures proper DNA replication origin firing, critical for genome stability and normal cell cycle progression. Defects or silencing of OBI1 result in impaired replication origin firing, reduced replisome formation, and potentially altered cell proliferation; it is implicated in cancer biology, and its molecular activity may represent a potential therapeutic target for diseases involving dysregulated DNA replication.
