Overview

Origin recognition complex subunit 1 (ORC1) is the largest subunit of the origin recognition complex, essential for the initiation of DNA replication in eukaryotes. It binds origins of replication as part of a heterohexameric complex, serving as a platform for pre-replicative complex assembly including Cdc6 and MCM proteins. ORC1's cell cycle-dependent regulation is mediated by transcriptional and proteolytic mechanisms. ORC1 is implicated in chromatin binding, centrosome and centriole copy number regulation, and transcription silencing through interaction with histone acetyltransferases. Clinically, ORC1 is overexpressed in several cancers, correlating with poor prognosis and suggesting utility as a diagnostic biomarker and potential therapeutic target. ORC1 pseudogene 1 (ORC1P1) is a noncoding pseudogene related to ORC1. No functional protein, biological activity, or relevance as a molecular target or biomarker has been described in available literature. ORC1P1 is not a canonical, functional molecular target.
