The organic matrix components of enamel and dentin are a specialized group of extracellular matrix proteins and proteoglycans that regulate the formation and structural integrity of teeth. In enamel, the matrix is primarily composed of amelogenin, enamelin, and ameloblastin, which guide the growth of hydroxyapatite crystals during amelogenesis. In dentin, the matrix consists largely of Type I collagen and non-collagenous proteins such as dentin sialophosphoprotein (DSPP) and dentin matrix protein 1 (DMP1), which provide a framework for mineralization and mechanical resilience. These components are essential for tooth development, and mutations in the genes encoding them lead to disorders like amelogenesis imperfecta and dentinogenesis imperfecta. From a therapeutic perspective, these matrix components are targeted or utilized in regenerative dentistry; for instance, enamel matrix derivatives (EMD) are used to promote periodontal tissue regeneration by mimicking the natural environment of tooth development. Additionally, biomimetic peptides and remineralizing agents interact with these organic structures to treat dental caries and dentin hypersensitivity. The matrix also plays a role in the mechanical toughening of mature enamel, and damage to these proteins can compromise tooth durability.
