Organophosphorus hydrolase (OPH), also known as phosphotriesterase (PTE), is a zinc-dependent enzyme found principally in soil bacteria such as Brevundimonas diminuta and Flavobacterium species. It belongs to the amidohydrolase superfamily and catalyzes the hydrolysis of a wide range of organophosphorus compounds, including many pesticides and chemical warfare agents. OPH has been engineered and explored as a therapeutic agent to detoxify organophosphate poisoning and is used in bioremediation applications for environmental clean-up. Structural and mechanistic studies reveal an (α/β)₈-barrel fold and a binuclear metal center essential for catalysis.
