Origin recognition complex subunit 4 (ORC4) is a protein that forms part of the six-subunit origin recognition complex (ORC), which is essential for the initiation of DNA replication in eukaryotic cells.[1][3][5][6][7][8] ORC4, along with the other five ORC subunits (ORC1-6), binds to specific DNA sequences at origins of replication, serving as a critical platform for recruitment of additional factors (such as CDC6, MCM2-7, and CDT1) that assemble the pre-replicative complex, allowing DNA synthesis to begin.[5][6][7][8] ORC4 contains an ATPase domain (AAA+ ATPase), and its ATP-binding motifs are crucial for replication activity. Mutations in ORC4 can disrupt replication licensing and are implicated in rare genetic diseases such as Meier-Gorlin syndrome, characterized by growth and developmental defects.[3][5][8] ORC4 does not currently represent a druggable therapeutic target, and no approved drugs directly interact with this protein.
