Overview

Origin recognition complex subunit 6 (ORC6) is the sixth and smallest subunit of the origin recognition complex (ORC), a highly conserved six-protein complex essential for the initiation of DNA replication in eukaryotic cells[2][3][4]. ORC6 is encoded by the *ORC6* gene (also called *ORC6L*) and plays a key DNA-binding role, anchoring the ORC to replication origins and facilitating pre-replication complex assembly[1][6]. Structurally, human ORC6 contains three independent domains and a distinct DNA-binding domain[4]. Functionally, ORC6 is essential for viability and proper chromosome replication and segregation; it also plays a role in cytokinesis, sometimes independently of the ORC[2][4][5]. In humans, mutations can result in syndromes characterized by developmental defects, including Meier-Gorlin syndrome[4]. ORC6 is not a typical therapeutic target, with no direct drugs acting upon it, but it is crucial for understanding DNA replication and cell division biology.
