Ornithine decarboxylase 1 (ODC1) is a gene encoding the enzyme ornithine decarboxylase, which catalyzes the decarboxylation of ornithine to putrescine, the first and rate-limiting step in polyamine biosynthesis. Polyamines are essential for cell growth and proliferation. ODC1 is notable for its regulation by antizyme and ubiquitin-independent proteasomal degradation. Dysregulation of ODC/ODC1 has been linked with various cancers and neurodevelopmental disorders. The enzyme's activity can be specifically inhibited by compounds such as DL-alpha-difluoromethylornithine (DFMO), which has been used experimentally both as an anti-cancer agent and as a tool for studying developmental biology.
