The Ornithine decarboxylase antizyme 1 (OAZ1) regulatory RNA is a specialized structural element within the OAZ1 mRNA that acts as a sensor for intracellular polyamine levels (Matsufuji et al., 1995, Cell). It contains a unique +1 ribosomal frameshift site and a downstream pseudoknot that are essential for the translation of the functional antizyme 1 protein (Ivanov et al., 2000, Nucleic Acids Res). When polyamine concentrations (such as spermidine and spermine) are high, they interact with the ribosome and the RNA structure to induce a frameshift, allowing the production of OAZ1. OAZ1 then binds to ornithine decarboxylase (ODC), the rate-limiting enzyme in polyamine biosynthesis, and targets it for ubiquitin-independent degradation by the 26S proteasome (Pegg, 2006, Am J Physiol Cell Physiol). This feedback mechanism is vital for maintaining polyamine homeostasis, as elevated polyamines are linked to rapid cell proliferation and oncogenesis. Consequently, this regulatory RNA is a target for polyamine mimetics and small molecules designed to suppress ODC activity in cancer therapy (UniProt P54368).
