Orthopedia homeobox (OTP) is a highly conserved homeodomain-containing transcription factor involved in the development of the central nervous system across species[1][2][3][4][5]. In humans, OTP is encoded on chromosome 5q14.1 and is expressed predominantly in restricted regions of the developing brain, particularly contributing to hypothalamic neuroendocrine systems[1][2][3][5]. Its activity specifies regional identities during early neural and gut development and is essential for differentiation of key neuroendocrine hypothalamic cell types[1][3]. OTP is currently under study as a prognostic biomarker for lung carcinoid tumors and its function in human tumor biology is being actively explored[2][5]. Disruption of OTP leads to severe neurodevelopmental phenotypes in animal models[3]. OTP does not currently have known small molecule agonists or antagonists, and it is not established as a drug target in pharmacology[2][4][5]. Its primary clinical significance relates to its potential as a diagnostic or prognostic marker in specific neuroendocrine tumors[5].
