Overview

The Orthopoxvirus VP37 envelope wrapping protein, also known as P37 or F13 (gene F13L in vaccinia virus), is a highly conserved viral protein found in all members of the orthopoxvirus genus, including variola (smallpox), monkeypox, cowpox, and vaccinia viruses. It is essential for the formation of the viral envelope that allows mature virions to exit infected cells and spread within the host. VP37 mediates envelopment of intracellular mature virus (IMV) particles, requiring interaction with host cell proteins Rab9 GTPase and TIP47 for virion egress. Tecovirimat is an FDA-approved antiviral drug that specifically targets VP37, blocking its interaction with Rab9/TIP47 and preventing viral spread.
