Osteogenesis-associated long non-coding RNA (lncRNA-OG) is a long intergenic non-protein coding RNA upregulated during the osteogenic differentiation of bone marrow-derived mesenchymal stem cells[1]. It acts epigenetically and through direct protein interactions (notably with hnRNPK) to regulate the expression of bone morphogenetic protein (BMP) family genes, particularly impacting the BMP/Smad1/5/8 signaling pathway. Its expression promotes differentiation of stem cells into osteoblasts—a major cellular component in bone formation—while knockdown inhibits this process. It operates mechanistically through modulation of promoter acetylation (H3K27ac), serving as a critical regulator of cellular fate in bone tissue. As such, it represents a novel therapeutic target for promoting bone regeneration, though no approved drugs yet directly target this lncRNA[1].
