Osteogenic cells, primarily comprising osteoprogenitor cells and osteoblasts, are the essential cellular components responsible for bone matrix synthesis and mineralization at bone defect sites (StatPearls, 2023). These cells are derived from mesenchymal stem cells and are regulated by a variety of systemic hormones and local growth factors, such as Bone Morphogenetic Proteins (BMPs) and Wnt signaling molecules (PubMed, PMID: 30233447). In therapeutic contexts, bone defect sites are targeted to promote osteogenesis through the use of osteoinductive agents, osteoconductive scaffolds, or cell-based therapies (NIH, 2022). For instance, recombinant human BMP-2 is used to induce the differentiation of these cells to bridge critical-sized defects. Understanding the recruitment and activation of osteogenic cells is crucial for treating complex fractures, non-unions, and bone loss resulting from trauma or tumor resection (PubMed, PMID: 31433430).
Therapeutic agents target osteogenic cells by activating signaling pathways such as BMP/Smad and Wnt/beta-catenin, which promote the differentiation of mesenchymal stem cells into mature osteoblasts that synthesize and mineralize the bone matrix (PubMed, PMID: 28838613).
