Osteoprogenitor cells are specialized mesenchymal stem cell-derived precursors committed to the osteoblast lineage, playing a fundamental role in bone formation and maintenance (StatPearls, NBK541037). These cells are primarily located in the periosteum and endosteum, where they serve as a reservoir for new osteoblasts during growth and fracture repair (Bone Research, 2020). Their differentiation is tightly regulated by key transcription factors such as Runx2 and Osterix, as well as signaling pathways including Wnt/beta-catenin and Bone Morphogenetic Protein (BMP) signaling (PubMed, 10409725). In clinical practice, these cells are the primary focus of anabolic therapies for osteoporosis, where drugs like Teriparatide stimulate their proliferation and maturation to increase bone mass (NEJM, 2001). Dysregulation or depletion of the osteoprogenitor pool is associated with age-related bone loss and impaired healing of skeletal injuries (Nature Reviews Endocrinology, 2021).
