Overview

OSTM1 antisense RNA 1 (OSTM1-AS1) is a long noncoding RNA that acts as an oncogenic factor in Wilms' tumor by sponging miR-514a-3p, which relieves microRNA-mediated suppression of MELK, thereby driving tumor cell proliferation, migration, and resistance to apoptosis. Its expression is significantly increased in Wilms' tumor tissues, and genetic knockdown experiments demonstrate reduced cell growth and tumorigenicity, suggesting its potential as a biomarker and therapeutic target for Wilms' tumor. OSTM1-AS1 exemplifies the disease relevance of antisense long noncoding RNAs in cancer biology and post-transcriptional gene regulation.

Mechanism of action

Inhibition via antisense RNA knockdown (e.g., siRNA or antisense oligonucleotides theoretically could reduce OSTM1-AS1 expression, leading to decreased tumor cell viability and migration). Additionally, it functions through miRNA pathway modulation by sponging miR-514a-3p to relieve suppression of MELK, a kinase involved in cell proliferation.
