These interactions typically occur in the 3' untranslated regions (UTRs) of the off-target mRNAs and lead to their degradation or translational repression, mimicking the mechanism of endogenous microRNAs (Birmingham et al., 2006, Nature Methods). In the context of ITGB4, which encodes a subunit of the alpha-6 beta-4 integrin receptor involved in cell-matrix adhesion and signaling (UniProt P16144), off-target effects can complicate the interpretation of functional studies and pose safety risks in therapeutic applications. Managing these effects requires careful siRNA sequence design, chemical modifications such as 2'-O-methyl groups, and rigorous validation using multiple siRNA sequences or rescue experiments (Fedorov et al., 2006, Nucleic Acids Research).
