Other mRNAs with partial complementarity to the ITGB4 siRNA guide strand refers to the unintended transcript targets of a small interfering RNA (siRNA) designed to silence the ITGB4 gene (Integrin subunit beta 4). This phenomenon, known as off-target silencing, occurs when the siRNA guide strand binds to non-target mRNAs through partial sequence homology, most commonly involving the 'seed' region (nucleotides 2-8) (Jackson et al., 2003, Nature Biotechnology). In the context of ITGB4, which encodes a subunit of the alpha-6 beta-4 integrin receptor involved in cell-matrix adhesion and signaling (UniProt P16144), off-target effects can complicate the interpretation of functional studies and pose safety risks in therapeutic applications. Managing these effects requires careful siRNA sequence design, chemical modifications such as 2'-O-methyl groups, and rigorous validation using multiple siRNA sequences or rescue experiments (Fedorov et al., 2006, Nucleic Acids Research).
