Overview

Other non-olfactory G protein-coupled receptors is a broad classification used to group GPCRs that are not involved in sensory perception (olfaction) and do not belong to the primary, well-defined families such as Rhodopsin, Secretin, or Glutamate receptors (Alexander et al., 2023). This category frequently includes orphan receptors, for which the endogenous ligand has not yet been identified, as well as miscellaneous Class A receptors that exhibit unique structural or functional characteristics (Hauser et al., 2017). These receptors play critical roles in diverse physiological processes, including metabolic homeostasis, immune response, and central nervous system signaling (Insel et al., 2019). Because many members of this group are expressed in a tissue-specific manner and are implicated in diseases like cancer and diabetes, they are highly valued in drug discovery as potential novel therapeutic targets (Gaulton et al., 2017). However, as a collective term rather than a single molecular entity, it encompasses a wide variety of mechanisms of action and pharmacological profiles depending on the specific receptor being addressed.
