Overview

The "Other protein kinases" group is a classification of approximately 83 human kinases that do not fit into the seven major established kinase families (Manning et al., 2002). This group is highly diverse and includes essential mitotic regulators such as Aurora kinases (AURKA, AURKB) and Polo-like kinases (PLK1-4), as well as kinases involved in the DNA damage response like WEE1 (UniProt, 2024). Because these enzymes are often overexpressed in malignant cells to drive uncontrolled proliferation, they have become significant targets for anti-cancer drug development (Nature Reviews Drug Discovery, 2018). Therapeutic agents targeting this group, such as alisertib and adavosertib, generally function as small-molecule inhibitors that block the phosphorylation of downstream substrates (PubMed, 2021). However, since this is a broad category rather than a single target, the clinical utility and safety profile of any given inhibitor depend entirely on the specific kinase member it targets and its selectivity across the kinome (NIH, 2023).
