Otoconin-90 (OC90) is a glycoprotein comprising the vast majority of the protein content in otoconia of mammals, which are calcium carbonate crystals crucial for sensing gravity and balance in the vestibular system[1][3][4]. OC90 has two domains homologous to secretory phospholipase A2, contributing to its function in matrix organization and crystal formation, although it lacks enzymatic activity and instead acts structurally to organize and scaffold crystal nucleation and growth[1][3]. It interacts with other matrix proteins like Otolin-1, ensuring proper organization and size of otoconia[4]. OC90 deficiency leads to impaired otoconia biogenesis and severe vestibular dysfunction, as demonstrated in knockout mouse studies[4]. Because OC90 is not an enzyme, receptor, or transporter, it is not considered a conventional pharmacological target. There are no known drugs or compounds that interact directly with OC90 for therapeutic purposes[1][3][4].
