Overview

OTU domain-containing protein 5 (OTUD5, also known as DUBA) is a deubiquitinating enzyme that modulates protein stability and signaling in immune cells. It directly regulates ubiquitination of key adaptor proteins (notably STING and TRAF3), promoting the stability of proteins involved in antiviral and antitumor immune responses or attenuating inflammatory signaling via TLR and RLR pathways. OTUD5 is essential for robust type I interferon signaling and can regulate T-cell responses, apoptosis via TP53, and the regulation of IL-17 production. While OTUD5 is recognized as a regulatory node in immunity and tumor biology, the “novel transcript, antisense to OTUD5” (ENSG00000303123) is not a validated molecular target and lacks functional annotations or clinical relevance in current databases.
