Overview

OTX2 antisense RNA 1 (OTX2-AS1) is a long non-coding RNA transcribed from the opposite strand of the OTX2 gene locus, regulated by a common bidirectional promoter with OTX2. It is non-protein coding and is mainly implicated in the regulation of gene expression possibly through RNA-RNA or RNA-protein interactions[6]. OTX2-AS1 is significantly upregulated in several cancer types, particularly medulloblastoma, where it promotes tumor cell growth, viability, and migration. Its expression is also associated with immune infiltration and poorer prognosis in various cancers, including breast cancer, lung squamous cell carcinoma, and gastric cancer[2][3][5]. Notably, OTX2-AS1 upregulation makes medulloblastoma cells more vulnerable to BCL-2 inhibition, suggesting indirect therapeutic implications[3].
