Outer dynein arm docking complex subunit 1 (ODAD1), also known as coiled-coil domain-containing protein 114 (CCDC114), is a structural protein crucial for the proper assembly and attachment of outer dynein arms to the doublet microtubules in the axonemes of eukaryotic cilia and flagella[7]. ODAD1 is a component of the outer dynein arm docking complex (ODA-DC), which typically consists of three subunits and is essential for ciliary motility[1][3][5]. The ODA-DC acts as an adaptor or scaffold, stabilizing the association of the outer dynein arms at regular 24-nanometer intervals along the axoneme, which is necessary for effective ciliary beating[1][3][5]. Mutations in ODAD1 can cause primary ciliary dyskinesia (PCD), a disorder characterized by impaired ciliary movement and its clinical manifestations such as chronic respiratory tract infections and reduced fertility[7]. There are currently no known drugs that directly target ODAD1, and it is not considered a therapeutic target for pharmacologic intervention. However, its genetic alteration serves as a diagnostic biomarker for certain ciliopathies, most prominently primary ciliary dyskinesia[7].
