Outer dynein arm docking complex subunit 4 (ODAD4; also known as TTC25 or Tetratricopeptide repeat protein 25) is a protein localized to ciliary axonemes and is crucial for the docking of outer dynein arms (ODA) onto cilia microtubules[2][5]. ODAD4 is a component of the heterotrimeric outer dynein arm-docking complex (ODA-DC), which organizes and binds ODA to the doublet microtubule every 24 nm, ensuring proper and efficient ciliary beating[3][4][6][7]. ODAD4 contains tetratricopeptide repeat (TPR) domains, which mediate protein-protein interactions important for ODA-DC assembly[2]. Mutations in ODAD4 disrupt ODA docking and ciliary motility, resulting in primary ciliary dyskinesia type 35, characterized by chronic respiratory infections, impaired sperm motility, and left-right body asymmetry defects such as situs inversus[2]. There is no evidence that ODAD4 is a therapeutic target or interacts with approved drugs[2][5].
