The outer membrane lipid bilayer is a specialized biological membrane found primarily in Gram-negative bacteria. It serves as a protective barrier and plays critical roles in cellular structure, nutrient acquisition, and defense against environmental threats such as antibiotics. The outer leaflet is dominated by glycolipids, especially lipopolysaccharide (LPS), which is responsible for the endotoxic effects associated with Gram-negative bacterial infections. The overall structure provides an asymmetric arrangement that is essential for its function and integrity. It acts as a selective permeability barrier and protects against external toxic threats including antibiotics by limiting their access to inner cellular components. LPS on the surface triggers strong immune responses in host organisms. The asymmetry between leaflets (phospholipid-rich inside, LPS-rich outside) is actively maintained. This unique composition distinguishes it from other biological membranes and underlies its critical role in bacterial physiology and pathogenicity.
