The ovarian stem cell niche is a specialized microenvironment within the ovary, primarily located in the hilum region and the junction of the ovarian surface epithelium (OSE) with the mesothelium, that regulates the maintenance, self-renewal, and differentiation of ovarian stem cells (Flesken-Nikitin et al., 2013, Nature). This niche is composed of a complex architecture including stromal cells, extracellular matrix components, and a specific milieu of signaling molecules such as Wnt, Notch, and Bone Morphogenetic Proteins (BMPs) (Ng and Surani, 2011, Science). Under normal physiological conditions, the niche facilitates the repair of the OSE after ovulation and supports the survival of very small embryonic-like stem cells (VSELs) or germline stem cells (Virant-Klun, 2015, Stem Cells International). However, the niche plays a critical role in disease, particularly in ovarian cancer, where it can be co-opted to form a "cancer stem cell niche" that protects malignant cells from chemotherapy and promotes metastasis (Ahmed et al., 2013, Journal of Hematology & Oncology). While the niche itself is not a single druggable molecule, therapeutic interventions often target its constituent pathways—such as using VEGF inhibitors like Bevacizumab to alter the vascular component or experimental Notch inhibitors to disrupt stem cell maintenance (Lau et al., 2008, Carcinogenesis). Understanding the niche's regulatory mechanisms is essential for developing strategies to regenerate ovarian function or to eradicate chemoresistant cancer cell populations.
Therapeutic strategies do not target the niche as a single entity but rather aim to disrupt the supportive signaling pathways (e.g., Notch, Wnt, Hedgehog) or the vascular and extracellular matrix components that constitute the microenvironment (Ahmed et al., 2013, Journal of Hematology & Oncology).
