OTUD6B-AS1 is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the OTUD6B gene on chromosome 8q21.3 (Zheng et al., 2025 [1]). It acts as a multifaceted regulator in various human malignancies and autoimmune conditions like systemic sclerosis, where it exhibits a dual role as either an oncogene or a tumor suppressor depending on the specific tissue context (Takata et al., 2019 [2]). The primary mechanism of action for OTUD6B-AS1 is its function as a competitive endogenous RNA (ceRNA), sponging microRNAs such as miR-3171 and miR-664b-3p to modulate downstream signaling pathways, most notably the Wnt/β-catenin and PI3K/AKT/mTOR pathways (Wang et al., 2021 [3]; Kong et al., 2020 [4]). In cancers like breast and hepatocellular carcinoma, its upregulation promotes tumor growth, metastasis, and resistance to therapies like paclitaxel and cisplatin (Li et al., 2021 [5]). Conversely, in colorectal and renal cell carcinomas, it often acts as a tumor suppressor, and its downregulation is associated with poor prognosis (Wang et al., 2019 [6]). Given its significant impact on cellular processes like ferroptosis, apoptosis, and epithelial-mesenchymal transition, OTUD6B-AS1 is being investigated as both a diagnostic biomarker and a potential therapeutic target for RNA-based interventions (GeneCards [7]). Research into targeting OTUD6B-AS1 using antisense oligonucleotides (ASOs) has shown promise in preclinical models for inhibiting tumor progression and modulating immune responses (Takata et al., 2019 [2]).
