Overview

Ovo-like transcriptional repressor 1 (OVOL1) is a conserved zinc finger transcription factor that modulates epithelial and germ cell differentiation by repressing genes that maintain progenitor states. It is essential for transitions from proliferation to terminal differentiation in epithelial tissues, including hair follicles, kidney, and placenta. OVOL1 acts through dual mechanisms: it competes with transcriptional activators for promoter binding and actively recruits histone deacetylases via its SNAG domain, resulting in chromatin condensation and gene silencing[1][3][4]. In cancer, OVOL1 suppresses epithelial–mesenchymal transition, inhibits cell migration, and stabilizes SMAD7, making it a prognostic marker and potential therapeutic target in epithelial-derived tumors such as breast cancer[5].
