Overview

Oxidative stress-responsive serine-rich protein 1 (OSER1) is a highly conserved, nuclear, serine-rich protein coded by the OSER1 gene (also known as C20orf111) in humans and homologous genes in many species. OSER1 is not a classical drug target (such as a receptor, kinase, or transporter) but is instead an evolutionarily conserved effector protein that is transcriptionally regulated by FOXO transcription factors. OSER1 has been shown to improve cellular and organismal resistance to oxidative stress by supporting mitochondrial integrity and lifespan in several species including nematodes, flies, and silkworms; its overexpression prolongs lifespan and increases tolerance to stressors, while depletion shortens lifespan and heightens vulnerability to oxidative damage. Protein expression is nuclear, and the protein contains a large DUF776 domain of unknown function, with extensive predicted serine phosphorylation sites. Polymorphisms in OSER1 are associated with human longevity, and there is evidence it may have roles in reproduction. Expression is increased in certain stress and apoptotic contexts (such as in response to hydrogen peroxide). The exact molecular and regulatory mechanisms remain incompletely understood, and no drugs directly target OSER1 as of current knowledge.
