Oxygen consumption refers to the physiological process and rate (OCR) at which cells utilize oxygen to produce adenosine triphosphate (ATP) through oxidative phosphorylation in the mitochondria. It is not a single molecular target like a receptor or enzyme but rather a complex metabolic readout reflecting the integrated activity of the electron transport chain (ETC) and the citric acid cycle. Measurement of oxygen consumption is a fundamental tool in bioenergetics used to assess mitochondrial health and cellular metabolic state (NIH, 2023). In drug development, changes in oxygen consumption are often monitored to evaluate drug-induced mitochondrial toxicity or the efficacy of metabolic modulators in diseases like cancer and diabetes. Dysregulation of this process is associated with mitochondrial myopathies, the metabolic reprogramming seen in tumor cells (Warburg effect), and various age-related neurodegenerative disorders (StatPearls, 2023).
Modulation of oxygen consumption occurs through the inhibition or uncoupling of the mitochondrial electron transport chain complexes (I-IV) and ATP synthase (Complex V).
