Oxysterol-binding protein-like 6 (OSBPL6), also known as ORP6, is a member of the oxysterol-binding protein family of intracellular lipid receptors[1][2][3]. OSBPL6 contains an N-terminal pleckstrin homology domain and a C-terminal oxysterol-binding domain, which enables it to bind cholesterol and possibly other phosphoinositides[1][3]. OSBPL6/ORP6 is predominantly localized to endolysosomal compartments and the endoplasmic reticulum and plays a role in regulating cholesterol and sterol transport between these compartments to the ER, thereby influencing cholesterol esterification and efflux, particularly in macrophages and hepatocytes[1]. Its hepatic expression correlates positively with plasma HDL cholesterol in humans, and genetic variants in OSBPL6 are associated with both cardiovascular disease and Alzheimer’s disease risk, though its exact disease mechanisms require further elucidation[1][2][3]. OSBPL6 is transcriptionally regulated by the LXR transcription factors in response to elevated cholesterol, and post-transcriptionally by miRNAs such as miR-33 and miR-27b, positioning it as a key factor in cholesterol homeostasis networks[1].
