Overview

Oxysterol-binding protein-related protein 11 (OSBPL11), also known as ORP11, is a member of the oxysterol-binding protein (OSBP) family of intracellular lipid receptor proteins[1][2][3][4][7]. It contains an N-terminal pleckstrin homology domain and a highly conserved C-terminal sterol-binding domain, and predominantly localizes to the Golgi complex and late endosomes, where it dimerizes with its homolog ORP9[1]. OSBPL11 functions as an intracellular lipid transporter and sensor, important for lipid trafficking and metabolic homeostasis, notably mediating the exchange of phosphatidylserine and phosphatidylinositol-4-phosphate at ER-Golgi membrane contact sites, a step critical for sphingomyelin synthesis[2][7]. It is upregulated during adipogenesis, with roles in regulating adipocyte differentiation and triglyceride storage, and has been implicated in metabolic syndrome and cardiovascular disease risk through its effects on cholesterol/glucose metabolism[1][2]. In cancer, particularly hepatocellular carcinoma, downregulation of OSBPL11 has been observed, pointing to a possible biomarker role in liver disease[1]. OSBPL11 is not a current therapeutic target (no known drug interaction or mechanism of pharmacological targeting), but its function in lipid metabolism places it at the interface of metabolic disease and oncology research[1][2][7].
