Oxysterol-binding protein-related protein 1A (OSBPL1A), also known as ORP1 or ORP1L, is a member of the oxysterol-binding protein (OSBP) family, which serves as intracellular lipid receptors and lipid transfer proteins[3][2]. OSBPL1A contains an N-terminal pleckstrin homology (PH) domain, ankyrin repeats, and a conserved C-terminal OSBP-like (ORD) sterol-binding domain[2]. It binds phospholipids (with strong affinity for phosphatidic acid and weak affinity for phosphatidylinositol 3-phosphate) and cholesterol. This protein stabilizes the active (GTP-bound) form of the small GTPase RAB7A on late endosomes/lysosomes and plays a critical role in regulating membrane trafficking and late endosomal/lysosomal dynamics[1][3][2]. OSBPL1A has been linked with cholesterol efflux, endosome positioning, and, through its interactions, can affect cellular processes such as cholesterol homeostasis and vesicular transport. Disease associations include a link to esophageal adenosquamous carcinoma[3]. There are no reported small-molecule drugs or direct pharmacological agents known to target OSBPL1A as of this time.
