P antigen family member 5 (PAGE5) is a member of the cancer/testis antigen family, a group of proteins typically restricted in expression to the testis in normal adult tissues but aberrantly expressed in various tumors, including sarcoma and melanoma[5][11][12]. PAGE5 is an intrinsically disordered, small protein with largely unknown normal biological function; it has been linked to protection against programmed cell death (anti-apoptotic effects) and may facilitate cancer cell proliferation and resistance to cell death in tumors expressing this antigen[5][12]. The specific role of PAGE5 in cancer biology remains under investigation, but its highly restricted expression pattern and immunogenicity make it a potential candidate for immunotherapeutic strategies such as cancer vaccines and adoptive T-cell therapy[4][10]. There are no approved drugs directly targeting PAGE5; its value as a biomarker or therapeutic target is under ongoing research, with notable challenges including heterogeneous tumor expression and the risk of immune-related effects on normal testis tissue[4][6][12].
