p-Methylbenzoic acid, commonly known as p-toluic acid, is an aromatic carboxylic acid that serves as a significant intermediate in industrial chemical synthesis and a key metabolic product of p-xylene. In biological systems, it is formed through the oxidation of the methyl group of p-xylene, a process mediated by hepatic enzymes such as cytochrome P450, alcohol dehydrogenase, and aldehyde dehydrogenase. While it is a naturally occurring metabolite during the detoxification of environmental pollutants, it does not function as a therapeutic target for drug development. Instead, its presence in biological fluids, often as a glycine conjugate, is used as a biomarker for monitoring human exposure to volatile organic compounds like xylene. Industrially, p-methylbenzoic acid is a precursor in the production of terephthalic acid, which is essential for the manufacturing of polyethylene terephthalate (PET) plastics and fibers.
