The p14ARF-derived peptide ARF35-43 presented by HLA-A*02:01 is a specific peptide-major histocompatibility complex (pMHC) that serves as a tumor-associated antigen (TAA) for cancer immunotherapy. p14ARF is a tumor suppressor protein encoded by the CDKN2A locus that normally functions to stabilize p53 by inhibiting MDM2 [UniProt, P42771]. In many cancers, particularly those with p53 mutations or oncogenic stress from MYC/RAS signaling, p14ARF is paradoxically overexpressed because the normal feedback loops are disrupted [PubMed, 12738670]. This high intracellular concentration allows the ARF35-43 peptide (sequence: FVRSRVPAA) to be processed and presented on the cell surface by the HLA-A*02:01 allele. Therapeutic strategies, such as TCR-engineered T-cell therapies like MDG1015, are designed to recognize this specific pMHC complex with high affinity to induce targeted tumor cell lysis [Medigene, 2023]. Because p14ARF expression is negligible in most healthy adult tissues, this complex provides a selective window for treating various solid tumors while minimizing off-target effects.
