Overview

PAK1 is a serine/threonine protein kinase and a member of the p21-activated kinases (PAKs), which are critical effectors linking Rho family GTPases—specifically Cdc42 and Rac—to cytoskeletal reorganization and nuclear signaling. It plays essential roles in regulating cell motility, morphology, gene expression, cell cycle progression, survival, angiogenesis, DNA repair, hormone sensitivity, and metabolism. Mutations or dysregulation of PAK1 have been linked with several diseases, including intellectual developmental disorder, breast cancer, and acute myeloid leukemia. It is also implicated in oncogenesis generally, viral pathogenesis, cardiovascular dysfunctions, and neurological disorders.
