Overview

P21-activated protein kinase-interacting protein 1 (PAK1IP1) is a nucleolar WD-repeat protein involved primarily in the negative regulation of PAK1 kinase activity and the assembly of the ribosomal large subunit. It binds the N-terminal region of PAK1, inhibiting its activation by small GTPases such as CDC42. PAK1IP1 also regulates TP53-mediated cell cycle progression, notably inducing G1 cell cycle arrest and modulating nucleolar stress responses by interacting with MDM2 and affecting ribosomal protein dynamics. Mouse models indicate that loss of PAK1IP1 function leads to cell cycle arrest, impaired cranial neural crest cell survival, reduced protein synthesis, and delayed craniofacial skeletal formation. Its role in human disease is still under study, with associations to intellectual disability and certain cancers, but it is not a conventional drug target or receptor.
