Overview

P2Y receptors are a family of G protein-coupled receptors (GPCRs) that respond to extracellular purine and pyrimidine nucleotides, including ATP, ADP, UTP, and UDP (Abbracchio et al., 2006). These receptors are widely expressed across various tissues and play fundamental roles in physiological processes such as platelet aggregation, vascular smooth muscle contraction, and immune cell activation (Burnstock, 2018). In the specific context of dU937 cells—a differentiated human monocytic cell line—P2Y receptors like P2Y1, P2Y2, and P2Y6 are studied for their roles in macrophage-like signaling and inflammatory cytokine production (von Kügelgen, 2021). Therapeutically, P2Y receptors are major targets in cardiovascular medicine, where P2Y12 antagonists are used to prevent thrombotic events (PubMed). Ongoing research also explores their potential in treating inflammatory disorders, dry eye disease, and certain cancers due to their involvement in cell proliferation and immune modulation (StatPearls).
