P2Y purinoceptor 4 (commonly abbreviated as P2Y4) is a member of the P2Y family of G protein-coupled receptors that mediate cellular responses to extracellular nucleotides such as UTP. P2Y4 is expressed in various tissues, notably regulating signal transduction, cell proliferation, differentiation, and immune responses. It is involved in bone biology—participating both in anabolic and catabolic processes—as well as in inflammation and potentially neurological and cardiovascular disorders. Although no approved drugs specifically target P2Y4, it is considered a therapeutic target, especially for diseases related to disturbances in purinergic signaling[3].
Agonists (e.g., UTP) bind and activate the receptor, initiating G protein-coupled signaling cascades—typically increasing intracellular second messengers such as calcium[3]. Antagonists inhibit the receptor, thereby reducing downstream signaling[3].
