The P2Y12 purinergic receptor is a G protein-coupled receptor primarily activated by adenosine diphosphate (ADP). It plays a crucial role in platelet aggregation and microglial function. In platelets, it is essential for ADP-induced platelet activation and thrombus formation, making it a key target for antiplatelet drugs. Within the central nervous system, it is exclusively expressed on microglia, where it participates in physiological monitoring of neuronal function and microglial neuroprotection. Genetic polymorphisms in the P2RY12 gene have been associated with altered risk for various diseases, including epilepsy, acute myocardial infarction, and bleeding disorders.
