Overview

p38 beta mitogen-activated protein kinase (p38β, also called MAPK11) is a member of the p38 MAP kinase family of serine/threonine kinases, which are activated in response to environmental stresses and pro-inflammatory cytokines. p38β is structurally related to other p38 isoforms (p38α, p38γ, p38δ), sharing a conserved dual phosphorylation motif (Thr-Gly-Tyr) in its activation loop. p38β functions as a crucial intracellular signaling mediator, regulating processes such as inflammation, cell differentiation, cell cycle control, apoptosis, and response to cellular stress. It is activated by upstream kinases (mainly MKK3 and MKK6), leading to phosphorylation of various transcription factors and effector proteins, and is implicated in disease states including acute and chronic inflammation, cancer, and neurodegeneration. Drugs targeting p38 MAPKs, including p38β, have been explored for inflammatory diseases and cancer, but their clinical use is limited by challenges with specificity and safety profiles.
