Overview

p38 mitogen-activated protein kinases (p38 MAPKs) are a class of serine/threonine protein kinases that play a central role in cellular responses to stress and inflammation. They are activated by various extracellular stimuli and regulate processes such as cell differentiation, apoptosis, autophagy, and the production of inflammatory mediators. There are four main isoforms of p38 MAPK in mammals: p38α, p38β, p38γ, and p38δ. Dysregulation or persistent activation/inhibition is implicated in several diseases including cancer, neurodegenerative disorders, inflammatory diseases, and cardiovascular conditions. Numerous small-molecule inhibitors targeting different subtypes have been developed.
