Overview

p38α, also known as MAPK14, is a serine/threonine protein kinase and a member of the mitogen-activated protein kinase (MAPK) family. It plays central roles in cellular responses to stress, inflammatory signaling, cell survival/apoptosis, proliferation/differentiation/migration, and mRNA stability. It is activated by dual phosphorylation at the TGY motif by upstream kinases. p38α has complex roles in disease, acting as a tumor suppressor during early tumorigenesis but potentially promoting tumor progression at later stages or under certain conditions. Current inhibitors often lack selectivity, posing a challenge for therapeutic development.
