PICART1 is a long non-coding RNA (lncRNA) of 2.53 kb, encoded in the human genome at 17q21.33. It is transcriptionally upregulated by the tumor suppressor p53 and acts as a tumor suppressor itself by inhibiting cancer cell proliferation, migration, and invasion through suppression of AKT/GSK3β/β-catenin and JAK2/STAT3 signaling pathways. In several cancers such as breast, colorectal, and lung cancer, PICART1 expression is downregulated, and experimental overexpression of PICART1 can inhibit tumor growth and promote apoptosis. Its reduced expression also correlates with activation of immune pathways, and it is studied as a diagnostic or prognostic marker for cancers and immune-mediated neuropathies (AIDP, CIDP). PICART1 represents an emerging therapeutic and diagnostic RNA target, though no direct targeting drugs are currently approved.
Drugs/agents that increase p53 activity (such as RITA, doxorubicin) may upregulate PICART1; this leads to suppression of oncogenic signaling pathways (AKT/GSK3β/β-catenin, JAK2/STAT3) and tumor suppression.
