Overview

The p53-MDM2 axis is a critical regulatory pathway in cell biology, particularly in the context of tumor suppression and cancer development. It centers on the interaction between the tumor suppressor protein p53 and its primary negative regulator, MDM2. p53 is a transcription factor that responds to cellular stress by inducing cell cycle arrest, apoptosis, and DNA repair. MDM2 is an E3 ubiquitin ligase that binds to p53, inhibiting its transcriptional activity and promoting its degradation. The MDM2 gene is a direct transcriptional target of p53, forming an autoregulatory feedback loop. Dysregulation of this axis is a common event in cancer.
