A p53 response element (p53RE) is a specific DNA sequence in gene promoters or enhancers to which the p53 protein binds as a tetramer to modulate transcription of target genes, typically those involved in cell cycle arrest, DNA repair, apoptosis, and senescence[3][5][7][9]. The canonical consensus sequence is a tandem arrangement of two decameric half-sites, with the general formula RRRCWWGYYY (R = A/G, W = A/T, Y = C/T), sometimes separated by spacers of varying length[5][6][9]. Binding of p53 to these elements triggers activation or repression of gene expression in response to cellular stress, thereby safeguarding genomic stability and preventing tumorigenesis[1][3][5][9]. Unlike proteins or receptors, p53REs themselves are not the direct subject of pharmacological intervention; rather, they are an essential component of the cellular network through which p53 exerts critical tumor suppressor functions[2].
